Chemiluminescence immunoassay technique

Chemiluminescence immunoassay (CLIA) is a combination of chemiluminescence assay technique with high sensitivity and immunoassay reaction with high specificity. It is then used as an analytical technique for the detection of various antigens, semi-antigens, antibodies, hormones, enzymes, fatty acids, vitamins and drugs.

Chemiluminescence immunoassay is a new immunoassay technique developed after radioimmunoassay, enzyme immunoassay, fluorescence immunoassay and colloidal time-resolved fluorescence immunoassay.

Principle of chemiluminescence immunoassay technique

There are two parts in chemiluminescence immunoassay. That is, the immunoreaction technique and the chemiluminescence technique. The basic principle is that the enzyme in the immunoreaction acts on the luminescent substrate, causing a chemical reaction and releasing a large amount of energy to produce an excited state intermediate.

This excited state intermediates back to the stable ground state, can be emitted at the same time photons. Using the luminescence signal measurement instrument can measure the amount of light quantum yield. The photon yield is proportional to the amount of the substance to be measured in the sample, so that a standard curve can be established and the amount of the substance to be measured in the sample can be calculated.

According to the reaction mode, the chemiluminescence immunoassay technique is divided into double antibody sandwich, competition and indirect reaction modes.
